  • Research summary

Researchers are looking for a better way to treat advanced or metastatic TROP2-positive, non-squamous non-small cell lung cancer (NS-NSCLC). In NS-NSCLC the body is not able to control the growth of some cells in the lungs, these cells can form tumours. Advanced means treatment is difficult because the cancer has grown. Metastatic means the cancer has spread to other parts of the body. TROP2-positive means there are high levels of a protein called TROP2 on the surface of the cancer cells.

There are limited treatment options for people with this disease. Docetaxel is approved, but alone may not work well for everyone.

The trial drug, Dato-DXd, is designed to kill cancer cells that have TROP2. Researchers think Dato-DXd may work better than docetaxel in people with TROP2-positive cancer. This trial aims to learn how safe Dato-DXd is and how well it works compared to docetaxel in people with TROP2-positive disease.

This is a Phase 3, randomized, open-label trial of around 400 participants. Randomized means that treatment is randomly assigned to participants. Open-label means everyone involved will know what each participant receives. Participants will continue treatment unless their cancer gets worse, they have significant medical problems, or they leave the trial for other reasons. The trial will run at several National Health Service (NHS) hospitals in the UK.

Dato-DXd and docetaxel will be given by IV infusion every 3 weeks. Half of participants will receive Dato-DXd and half will receive docetaxel. Tests that will be performed include blood test, CT and/or MRI scan, vital sign measurements and heart and lung function tests among others.

A possible benefit for receiving Dato-DXd is improvement of advanced or metastatic NS-NSCLC however there are also a number of risks including inflammation of the lungs, eye problems, skin problems and mouth and throat problems, among others
