Research summary
Our research goal is to characterise T cell immune responses in individuals deemed to be at highest risk of developing rheumatoid arthritis (RA). To systematically document the evolution of responses requires analysis of T cells from patients both before, at disease onset and in established disease. Specifically we seek to understand what protein antigens derived from host tissues are responsible for stimulating arthritis provoking T cells. To this end we will exploit cutting edge technologies to study T cells in blood, lymph node and synovial biopsies, including sequencing of antigen receptor genes, mass spectrometry to detect protein antigens and in depth immune phenotyping using multi-parameter flow cytometry and mass cytometry. We will first study samples from patients with established RA to refine laboratory protocols for sample acquisition, processing and laboratory analysis. Once these have been optimised, we will apply these protocols to patients presenting with RA for the first time (drug naïve), and then to subjects at risk of developing RA.
