  • Research summary

Povorcitinib (also known as INCB054707) is a novel, potent, oral, small-molecule, selective, JAK1 inhibitor under clinical evaluation in various inflammatory disorders. Janus kinase signaling regulates several proinflammatory pathways and has been well recognized as a key driver for many diseases; further, JAK inhibitors have been approved for various conditions, such as rheumatoid arthritis, atopic dermatitis, and alopecia areata.
This rollover study is designed to enroll participants who are tolerating the study drug and receiving clinical benefit from it as part of predetermined, Incyte-sponsored parent clinical trials of povorcitinib. The parent Protocols contain the relevant disease background information and rationale for use of povorcitinib in each respective indication.
The purpose of this study is to continue assessing long-term safety across inflammatory conditions in which povorcitinib is being evaluated. Eligible participants include those who have completed predetermined, Incyte-sponsored, povorcitinib clinical trials and who have obtained clinical benefit during the parent study.
The dose of povorcitinib in this rollover study will be the same dispensed to the participant during the period immediately preceding enrollment. If the dose in the parent study was blinded, participants should start the rollover study at the lowest povorcitinib dose utilized in the parent study for the respective indication.
This study will collect and assess safety information with regard to AEs; disease-specific efficacy and QoL outcomes may also be collected, depending on the indication.
