  • Research summary

Researchers are looking for a better way to treat solid tumours (a type of cancer that starts in an organ or tissue of the body).
Some solid tumours are homologous recombination repair (HRR)-defective, which means the cells cannot properly fix certain changes to the DNA in the HRR genes, which can allow the tumour to spread. HRR-defective cancer cells commonly start growing in the ovaries or lining of the uterus, prostate, breasts, or pancreas.
The trial drug AZD4956 is designed to stop cancer cells from growing by blocking a protein that HRR-defective cancer cells use to repair damaged DNA.
In this trial, researchers want to learn about the safety of AZD4956, how it moves through and affects the body and how well it works in adults with solid tumours.
Researchers want to learn more about AZD4956 taken alone and to learn more about AZD4956 taken with an anticancer drug called saruparib. Saruparib is designed to help stop tumour cells from repairing themselves when damaged. Different dose levels of AZD4956 will be tested to learn which dose is the best to use. AZD4956 and saruparib are taken as tablets by mouth. More modules with other treatments may be added to this trial in the future.
This is an open-label trial which means participants, researchers and trial doctors will know what medication each participant is taking. Participants will continue to receive treatment until their cancer gets worse, they have significant medical problems, or they leave the trial for another reason. This trial which will be the first time AZD4956 is given to people.
The trial will include up to 191 participants from about 6 countries around the world. To join this trial, people must be at least 18 and have advanced or metastatic solid tumours.
During the trial, the doctors will do the following: blood and urine tests, take a sample of some of the participants’ tumours, physical examinations, heart tests like ECGs, measure vital signs and perform CT, MRI, and/or bone scans.
