  • Research summary

A number of different treatments may be used in the treatment of systemic lupus erythematosus (SLE) but treatment responses to each vary substantially. There are no predictive markers or tests which indicate who is most likely to respond to a specific treatment and which treatment should be best selected for any single individual. This means some people with lupus go through many different unsuccessful treatments before their condition improves.

This study will use high-resolution technologoies to characterise the immune pathways, proteins and genes modified by specific SLE treatments and which associate with clinical improvement. This aims to establish measurable blood-based markers which predict treatment response.

Adult subjects with clinically active SLE who are starting a treatment with mycophenolate mofetil or belimumab within routine NHS Rheumatology services will be invited to participate. The characteristics, activity and quality of life impact of their condition will be evaluated before starting treatment with either mycophenolate mofetil or belimumab, and again to understand their response at 3 months and at 6 months on treatment. Blood samples donated at these time points will be processed for single cell sequencing and proteomics analysis to generate an in depth analysis of immune changes. Samples and data in this study will be analysed by investigators in a research consortium composed of Newcastle University, Brigham and Women’s Hospital and Harvard Medical School (Boston, USA), Johns Hopkins (Baltimore, USA), and UMass Chan Medical School (Massachusetts, USA) supported by the Lupus Research Alliance.
