Research summary
Fourteen frozen colon cancer and adjacent normal tissues are a gift from Professor Michael Linnebacher at the University of Rostock and provided to enable the study of tumour antigens in colorectal tissues. The tissues will be acquired under the auspices of an existing ethical approval – attached to this application (ethical statement number: II HV 43/2004 (Ethikkommission an der Medizinischen Fakultät der Universität Rostock; St.-Georg-Str. 108, 18055 Rostock). The existing ethical approval allows the collection and distribution of the tissues to collaborators of Professor Linnebacher.
The tissues, following LREC and University of Hull ethical approval, will be sliced and used to make multiple tissue arrays. The samples will be examined for the expression of proteins that may act as targets for immunotherapy, biomarkers for early diagnosis of colon cancer or help us better understand the biological process of tumour development. Once the tissues samples are sliced they will be analysed in the laboratory using the techniques of immunohistochemistry on frozen tissues, Western blotting and real time PCR (qPCR). The principal investigator, Dr Barbara Guinn and students under her direction, all of whom will have been trained in safe and appropriate handling of primary tissues and the analysis of proteins present therein. Samples will not be given to a third party without the agreement of the guardian of the samples, Professor Michael Linnebacher.
