Research summary
Heart Failure with Reduced Ejection Fraction (HFrEF) is a condition where the heart is weak and cannot pump blood as well as it should.
A protein called CaMKII (calcium/calmodulin-dependent protein kinase II) plays an important role in keeping calcium levels balanced within heart cells, which is essential for the heart to squeeze and relax normally. In HFrEF, CaMKII activity can become overactive, causing harmful changes in heart muscle cells and making heart function worse. The research medicine CRD-4730 is designed to block or hinder CaMKII activity in HFrEF and may help prevent or reverse this damage.
The purpose of this study is to learn about the effects of the research medicine CRD-4730, on heart function and to see how safe CRD-4730 is for patients with HFrEF.
This is a randomised trial being conducted at clinical centres around the world. Participants receive either CRD-4730 tablets or placebo tablets. Three different doses of CRD-4730 will be tested during the trial.
The trial will include around 525 participants who are between 18 and 85 years old, have diagnosis of HFrEF and have been on a stable dose of heart failure medicines for at least the last 4 weeks prior to entering the trial. This trial will not include participants who need to take certain medications or who have certain medical conditions that, in the opinion of the trial doctor, would make it unsafe for them to participate or difficult to complete the trial measurements.
In this study, CRD-4730 or placebo will be added to the heart medicines participants are already taking. Participants will be assigned randomly by chance (like drawing straws) to receive either low, medium or high dose of CRD-4730 or placebo. Participants will take two tablets of CRD-4730 or placebo twice a day. The study medicine will be taken orally (by mouth) every day during the 24 weeks of the study treatment period.
Participants will have regular visits with the trial doctor throughout the study.
